BaseScope™ VS Reagent Kit – RED

The BaseScope™ VS Reagent Kit is designed to be automated on the DISCOVERY ULTRA Automated IHC/ISH slide staining systems from Roche Tissue Diagnostics. The BaseScope Assay leverages the same platform as proven and established within the RNAscope™ technology but expands the detection capabilities, enabling the detection of short RNA target sequences and exon junctions in cells and tissues with morphological context, providing new data dimensions and unique insight into biological mechanisms.

BaseScope™ LS Reagent Kit – RED

The BaseScope LS Reagent Kit is designed to be automated on the Leica Biosystems BOND RX Research Advanced Staining System. The BaseScope Assay leverages the same platform as proven and established within the RNAscope technology but expands the detection capabilities, enabling the detection of short RNA target sequences and exon junctions in cells and tissues with morphological context, providing new data dimensions and unique insight into biological mechanisms.

The BaseScope LS Reagent Kit automated workflow requires:

  • BaseScope LS Target Probes (Catalog or Made-to-Order Probes)
  • BaseScope LS Control Probes (Select species-specific LS positive control probes or packs of probes and LS negative control probes)
  • BaseScope LS Reagent Kit
  • RNAscope Control Slides (optional)

 

RNAscope™ 2.5 LSx Reagent Kit-RED

Obtain exceptional sensitivity with RNAscope™ 2.5 LS Assay - now even faster with a simpler run set-up. Advanced Cell Diagnostics introduces RNAscope 2.5 LSx Reagent Kit - RED, our first pre-filled assay designed to be used as an automated assay on the Leica Biosystems' BOND RX Research Advanced Staining System.  With volumes to accommodate the processing of 60 slides, the RNA ISH reagents come pre-filled in Bond Rx containers fixed on two trays and include Fast Red chromogen, hematoxylin, bluing and protease.

Probe Compatibility

The RNAscope 2.5 LSx reagents utilize the existing RNAscope 2.5 LS Probes for optimal performance.

 

Simple and Rapid Run Set Up

The RNAscope 2.5 LSx Reagent Kit- RED with pre-assembled reagent trays significantly decreases the set up time at the bench and instrument. Furthermore, the barcoded containers enable accurate scanning and recording of reagents lot numbers. Run preparation is simple:

  1. Scan and register trays and reagents
  2. Establish run parameters
  3. Select staining protocol
  4. Start your run

RNAscope™ VS Duplex Assay

The RNAscope VS Duplex Assay is designed as an automated assay on the DISCOVERY ULTRA Automated IHC/ISH slide staining systems from Roche. The RNAscope VS Duplex Assay, is an RNA in situ hybridization assay based on ACD’s patented signal amplification and background suppression technology. The assay uses a novel and proprietary method of in situ hybridization (ISH) to visualize single RNA molecules per cell in a multitude of sample types mounted on slides. RNAscope Assays do not require the RNA-free environment used for traditional ISH.

RNAscope™ 2.5 LSx Reagent Kit-BROWN

Obtain exceptional sensitivity with RNAscope™ 2.5 LS Assay - now even faster with a simpler run set-up. RNAscope™ Control Slides for RNAscope™ and BaseScope™ Assays

Control slides are essential to verify the assay conditions. ACD strongly recommends that all new users begin their RNAscope™ or BaseScope™ Assay experiments by selecting human or mouse control slides and running the assay on these slides with positive and negative control probes. This first run would serve as an assay control check before performing runs using your target probes and samples. Please note that control slides for BaseScope™ and RNAscope™ are identical and named RNAscope™ Control Slides.

Two varieties of control slides are offered: human control slides containing FFPE cultured cell pellets of human HeLa cells and mouse control slides containing FFPE cultured cell pellets from mouse NIH 3T3 cells.

In addition to purchasing the control slides, you will need to purchase species-specific positive and negative control probes for either manual assays or automated assays.

Control Probes for Manual Assays - BaseScope™ RED

Species-specific positive and negative control BaseScope™ probes are needed to run with human or mouse control slides for technique quality control check and with your test sample for RNA quality control check.

ACD provides BaseScope™ positive control probes that target the PPIB gene and BaseScope™ negative control probes that target the bacterial DapB gene.

Pretreatment reagents are used for blocking endogenous peroxidase activity and to optimally permeabilize properly prepared samples. These reagents have been optimized to better access target RNA and reduce optimization time in multiple tissues such as formalin-fixed, paraffin-embedded (FFPE) (including archival) tissue, fresh‑frozen (FF) tissue, fixed‑frozen tissue, tissue microarray (TMA), and cell preparations. 

The BaseScope™ Reagent Kit v2-RED, is based on ACD’s patented signal amplification and background suppression technology. The assay uses a novel and proprietary method of in situ hybridization (ISH) to visualize single RNA molecules per cell in a multitude of sample types mounted on slides. The BaseScope Assay is used for the detection of short RNA targets (50-300 bases), highly homologous sequences, RNA mutations, and exon junctions for the detection of splice variants, circular RNA, and gene fusion.
