RNAscope™ Pretreatment Reagents

RNAscope™ Pretreatment reagents are used for blocking endogenous peroxidase activity and to optimally permeabilize properly prepared samples. These reagents have been optimized to better access target RNA and reduce optimization time in multiple tissues such as formalin-fixed, paraffin-embedded (FFPE) (including archival) tissue, fresh‑frozen (FF) tissue, fixed‑frozen tissue, tissue microarray (TMA), and cell preparations. The pretreatment reagents listed in this section are for the manual RNAscope™ and BaseScope™ Assays.

Control Probes for Manual Assays: RNAscope Multiplex Fluorescent Reagent Kit

Species-specific positive probes and negative control probes are needed to run with human or mouse control slides for uality control check of the technique and with your sample for the Sample or RNA quality control check. ACD provides positive control probes that target species-specific housekeeping genes and negative control probes that target the bacterial DapB gene.

When using this RNAscope Multiplex Fluorescent Reagent Kit with three target RNAs, you will work in channels 1, 2 and 3 so you will also need control probes for channel 1, channel 2 and channel 3. These species-specific positive control probes and negative control probes for channel 1 when processed with your sample will enable you to assess your sample/RNA quality and serve as control for your channel 1 target probe etc. ACD offers a 3-plex Negative Control Probe which contains probes for channel 1, 2 and 3. We also offer 3-plex RNAscope™ Positive Control Probes for Human, Mouse and Rat where POLR2A probe is available in channel 1 and PPIB probe in channel 2 and UBC probe in channel 3.

RNAscope™ 2.5 HD Assay - RED

The RNAscope 2.5 High Definition (HD)—Red Assay, is based on ACD’s patented signal amplification and background suppression technology. The assay uses a novel and proprietary method of in situ hybridization (ISH) to visualize single RNA molecules per cell in a multitude of sample types mounted on slides. RNAscope Assays do not require the RNA-free environment used for traditional ISH. The HD version is the most sensitive RNA ISH method and can be used for low expressing gene targets.

The RNAscope 2.5 HD Reagent Kit-RED assay with it Fast Red dye offers a higher contrast and is the first choice in situ hybridization applications where chromogenic staining with DAB is less desirable, such as staining of highly pigmented lung, liver, retina and melanoma tissue specimens. Also for genes where a lower expression is assumed ACD recommends this assay as the red dots clearly distinguish against the hematoxylin staining and are visible under are standard brightfield microscope.

VectaMount^TMpermanent Mounting Medium (Cat. No. 321584) is must-have accessory for the RNAscope™HD Duplex Assay (Cat. No. 322430).

Produced by Vector Laboratories, this mounting medium is an optically clear and odorless formula for permanently preserving histochemical stains or precipitable enzyme substrates in tissue sections or cell preparations. VectaMount™ Mounting Medium contains no toluene or xylene. It has a viscosity which provides for easy application and uniform spreading over the tissue section. Mounted sections are clear with an ideal refractive index suitable for high resolution oil immersion microscopy. VectaMount™ Mounting Medium preserves the color and intensity of preparations stained with enzyme substrates.

EcoMount^TM Mounting Medium (Cat.No. 320409) is a must have accessory for the RNAscope ^® 2.5 HD Assay-Red.

Produced by Biocare Medical, this polymer-based mounting medium does not contain hazardous reagents such as xylene, toluene or benzene as its base solvent. EcoMount permits long-term storage of slides with chromogens. EcoMount dries quickly and retains excellent refractivity.

ACD EZ-Batch™ Slide Processing System for Manual Assays

The ACD EZ-Batch™ Slide Processing System facilitates processing multiple sample slides simultaneously. We designed this product for higher efficiency in running the manual RNAscope™ assay protocol. The system comprises RNAscope™ EZ-Batch™ Slide Holder and EZBatch™ Wash Tray.

HybEZ™ II Hybridization System for Manual Assays

Successful implementation of RNAscope™ Assay is directly linked to hybridization environment. The ACD HybEZ™ II Hybridization System and its ability to accurately keep the temperature stable is essential to the success of RNAscope™ and BaseScope™ workflows.

The system comprises: HybEZ oven (PN 321710/321720), a humidity control tray (PN 310012), and HybEZ Humidifying Paper (2 sheets PN 310025), EZ-Batch Wash Tray (PN 321717), EZ-Batch Slide Holder (PN 321716)
