Cancer

Acidification of tumor at stromal boundaries drives transcriptome alterations associated with aggressive phenotypes.

Acidosis is a fundamental feature of the tumor microenvironment that directly regulates tumor cell invasion by affecting immune cell function, clonal cell evolution, and drug resistance. Despite the important association of tumor microenvironment acidosis with tumor cell invasion, relatively little is known regarding which areas within a tumor are acidic and how acidosis influences gene expression to promote invasion.

Time resolved Gene Expression Analysis during Tamoxifen adaption of MCF-7 cells identifies long non-coding RNAs with prognostic Impact.

Acquired tamoxifen resistance is a persistent problem for the treatment of estrogen receptor positive, premenopausal breast cancer patients and predictive biomarkers are still elusive. We here analyzed gene expression changes in a cellular model to identify early and late changes upon tamoxifen exposure and thereby novel prognostic biomarkers.

Long non‑coding RNA ENST00000547547 inhibits cell proliferation, invasion and migration in colorectal cancer cells.

Colorectal cancer (CRC) is one of the most common and lethal types of cancer worldwide. Multiple lines of evidence have illustrated that long non‑coding RNAs (lncRNAs) are critical molecules in the regulation of CRC development and progression. The identification of new lncRNAs is expected to provide new biomarkers and potential therapeutic targets for CRC diagnosis and prevention.

Members of the endocannabinoid system are distinctly regulated in inflammatory bowel disease and colorectal cancer.

Preclinical studies have demonstrated that the endocannabinoid system (ECS) plays an important role in the protection against intestinal inflammation and colorectal cancer (CRC); however, human data are scarce. We determined members of the ECS and related components of the 'endocannabinoidome' in patients with inflammatory bowel disease (IBD) and CRC, and compared them to control subjects.

Epithelial insulin receptor expression-prognostic relevance in colorectal cancer.

Abstract

BACKGROUND:

Metabolic reprogramming in cancer encompasses the insulin receptor (IR) as a player of energy homeostasis and proliferation. We aimed to characterize vascular (VIR) and epithelial (EIR) IR expression in CRC and correlate it with clinico-pathological parameters and survival.

METHODS:

LncRNA PCAT1 activates AKT and NF-κB signaling in castration-resistant prostate cancer by regulating the PHLPP/FKBP51/IKKα complex.

In PTEN-deficient prostate cancers, AKT signaling may be activated upon suppression of androgen receptor signaling. Activation of AKT as well as NF-κB signaling involves a key regulatory protein complex containing PHLPP, FKBP51 and IKKα. Here, we report a critical role of lncRNA PCAT1 in regulating the PHLPP/FKBP51/IKKα complex and progression of castration-resistant prostate cancer (CRPC).

Loss of BCL9/9l suppresses Wnt driven tumourigenesis in models that recapitulate human cancer

Different thresholds of Wnt signalling are thought to drive stem cell maintenance, regeneration, differentiation and cancer. However, the principle that oncogenic Wnt signalling could be specifically targeted remains controversial. Here we examine the requirement of BCL9/9l, constituents of the Wnt-enhanceosome, for intestinal transformation following loss of the tumour suppressor APC.

Circ-ZNF609 regulates G1-S progression in rhabdomyosarcoma.

Circular RNAs (circRNAs) represent a class of covalently closed RNAs, derived from non-canonical splicing events, which are expressed in all eukaryotes and often conserved among different species.
