
Webinar: Tissue Imaging of Oligonucleotides – Cell Type Specific Distribution and Knock-Down Analysis in The Same Tissue Section with RNAscope™ ISH Assays
RNAscope in situ hybridization (ISH) is widely used to detect mRNA in tissue sections and is often combined with one protein marker. The RNAscope™ Plus assay enables the detection of one siRNA/ASO, up to three mRNAs, and one protein for cell-specific analysis of biodistribution and target knock-down. Integrating this assay with the PhenoImagerHT imaging system (Akoya Biosciences) allows for advanced image analysis by utilizing the auto fluorescent properties of the tissue to train AI classifiers. Moreover, the latest introduction of a protease-free RNAscope workflow allows for the detection of 1 siRNA, upto 1 to 3 mRNAs, and 2-4 proteins in the same tissue section. This is achieved by sequential protein detection using the TSA-workflow (Akoya Biosciences), creating a powerful tool for tissue analysis of oligonucleotides.
