The stomach epithelium is capable of constant regeneration driven by long-lived resident stem cells, but the mechanism that regulates their identity and turnover is not well known.



In this webcast, the speakers will demonstrate that Axin2+/Lgr5- cells constitute a highly proliferative, non-differentiated compartment capable of regenerating entire antral glands and that Rspo3 is a critical niche-specific regulator that orchestrates stem cell homeostasis. The ability of stromal niche cells (Rspo3+) to control and adapt the dynamics of epithelial stem cells (Axin2+/Lgr5-) constitutes a sophisticated mechanism that orchestrates epithelial regeneration and maintenance of tissue integrity.



During this webcast you will learn how the RNAscope Dual ISH technology was utilized to thoroughly characterize the gastric antral gland stem cell niche.

  • R-spondin family members enhance Wnt signaling
  • Of the 4 Rspo members examined, only Rspo3 was expressed in the mouse antrum, specifically in the stroma adjacent to the stem cell compartment
  • Dual RNA ISH showed that Rspo3 is produced by alpha-smooth muscle actin (α-SMA)-positive myofibroblasts in lamina muscularis mucosa directly beneath the glands
  • The increase in stem cell turnover in response to the gastric pathogen H. pylori is driven by Rspo/Wnt signaling
