Examination of Microglia at Single Cell Resolution in Health and Disease

Outline:

Microglia are the brain's resident immune cells and play key roles in normal development, maintenance of homeostasis, and disease.  Using a combination of single cell sequencing and RNAscope we have uncovered novel populations of microglia in both early post-natal development, injury, aging, and neurodegeneration.  We have also found that microglia are uniquely sensitive cells and require great care when performing both scRNA-seq and smFISH/RNAscope to ensure accurate results.

Learning Objectives:

--Spatial Information from RNAscope is key component of single cell studies to both localize populations spatially and validate sequencing results.

--RNAscope verifies artifactual gene expression observed in scRNA-seq when cells isolated under improper conditions

--HiPlex assay enables significantly more accurate localization of specific cell states in situ
